Technology overview


Bicosome is a proprietary skin delivery technology focused on target delivery of active ingredients to specfic skin layers.

 

Bicosome is composed of a combination of two lipid assemblies. The first lipid assembly is forming so-called ‘smart discs’ that carry the active ingredients. A collection of smart discs is then enclosed by the second lipid assembly: an external lipid vesicle. The two lipid assemblies combined form a double encapsulation of the active ingredient.

 

Cryo-TEM image give a good visual overview of the Bicosome structure. Both the outer lipid assembly and the lipid structures that for the smart discs are clearly visible.

These cryo-TEM images show the resemblance of the Bicosome structure to the structure of the skin's natural lamelar bodies.

 

Upon application, the outer vesicle fuses with the skin's surface. The smart discs are thereby delivered to the skin. The discs then pass through the narrow inter-cellular spaces of the Stratum Corneum and start to penetrate into the skin.

When the targeted skin layer is reached, the smart discs become part of the skin's lipid structure and the active ingredient is released.

 

Smart discs are designed to reach specific skin layers and are thus able to deliver active ingredients to the skin layer where they need to be. Fluorescence microscopy images of skin samples treated with different Bicosome prototypes demonstrate the penetration of the smart discs to the targeted skin layers: Stratum Corneum, Epidermis and Dermis. The Bicosome prototypes used in these tests contain fluorescein as to visualize skin penetration.

Formulation guidelines


The following formulation guidelines will help in successfully using the Bicosome products in formulations:

 

Liquid (water-based) Bicosome® products:

  • Post-add Bicosome® to the O/W emulsion upon cooling down
  • Recommended dosage: 1 - 3%
  • Optimum pH range: 4 – 8
  • Preferred processing temperature: <50˚C

 

Lyophylized Bicosome® products (available for Bicosome® FS and Bicotene® Complex):

  • Post-add Bicosome® to the W/O emulsion upon cooling down
  • Recommended dosage: 0.2 – 0.6%
  • Preferred processing temperature: <50˚C

 

Dos and Don’ts:

  • Do not dissolve Bicosome® in any solvents
  • Avoid harsh surfactants
  • Do not use in formulations with alcohol content of >15%
  • Avoid strong oxidants, such as peroxides
